| Summary: | This work was to quantify the superficial growth and the pigmentation of filamentous fungi colonies by effect of an extremely low frequency oscillating magnetic field (OMF-ELF) depending on nutritional status of these microorganisms in two culture media. OMF-ELF of 2 mT by 60 Hz/200 V during 2 h (optimization of an experimental design) was applied at two filamentous fungi strains isolated from indoor environments: Aspergillus niger (hyaline mycelium seen under the optic microscope) and Cladosporium cladosporioides (pigmented mycelium), this latter strain was used as a negative control. The strains were inoculated in Petri dishes with Malt Extract Agar (MEA) and Czapek Dox Agar (CDA), incubated at 30°C and photographs were taken every 4 hours for 10 days. The area (superficial growth) and color intensity (pigmentation) of these colonies were evaluated by digital images processing (DIP) using MatLab®. The stimulating effect of OMF-ELF on the cellular metabolism of both fungal strains was demonstrated, with an increase of the superficial growth and a greater variation in pigmentation (indicative of sporulation by conidiogenesis) in CDA until reaching values of spores similar to when there are enough nutrients (MEA). The predictive mathematical model obtained by optimization of the experimental design to predict the growth of Aspergillus niger colony was validated with the algorithm implemented for the DIP. It was concluded that the OMF-ELF stimulated the metabolism of the studied fungal strains independently of the nutrients in the culture media and that the DIP is a precise technique to quantify the macroscopic phenomena that occur due to magnetobiological effects at the cellular level. El objetivo de este trabajo fue cuantificar el crecimiento superficial y la pigmentación de las colonias de hongos filamentosos por efecto del campo magnético oscilante de frecuencia extremadamente baja (CMO-FEB) en función del estado nutricional de estos microorganismos en dos medios de cultivo. Se aplicó CMO-FEB de 2 mT a partir de 60 Hz/200 V durante 2 h (condiciones obtenidas por optimización de un diseño experimental) a dos cepas de hongos filamentosos aisladas de ambientes interiores: Aspergillus niger (hifas hialinas) y Cladosporium cladosporioides (hifas pigmentadas vistas bajo el microscopio), usada esta última especie como control negativo. Las cepas inoculadas en placas de Petri de 90 mm con Agar Extracto de Malta (AEM) y Agar Czapek Dox (ACD), se incubaron a 30°C y se tomaron fotografías cada 4 horas por 10 días. Se evaluó el área (crecimiento superficial) y la intensidad del color (pigmentación) de estas colonias mediante procesamiento digital de las imágenes (PDI) empleando MatLab®. Se demostró el efecto estimulante del CMO-FEB sobre el metabolismo celular de ambas cepas fúngicas, con un aumento del crecimiento superficial y una mayor variación de la pigmentación (indicativo de esporulación por conidiogénesis) en ACD hasta lograr valores superiores en la cantidad de esporas similares a cuando existen suficientes nutrientes (AEM). Con el algoritmo implementado para el PDI se validó el modelo matemático predictivo obtenido por optimización del diseño experimental para predecir el crecimiento de la colonia de Aspergillus niger. Se concluye que el CMO-FEB estimuló el metabolismo de las cepas fúngicas estudiadas independientemente de los nutrientes del medio de cultivo y que el PDI es una técnica precisa para cuantificar los fenómenos macroscópicos que ocurren por efectos magnetobiológicos a nivel celular. |
