Sumario:Objective: The objective of this study was to analyze the postmortem gene expression by time-dependent observation. Design: The post-mortem spleen samples were taken from the mice sequentially and RNA analysis were performed for time-dependent observation. Materials and methods: Mice were killed by cervical spine subluxation with anesthetic treatment. Total RNA was extracted from incident spleen from mice and clarified the quality and quantity. To clarify the postmortem change, the differential display-PCR were performed and followed by cloning the different amplifications, DNA sequencing and homology searches, RI labelling the cloned fragments and northern blotting. Results: Postmortem RNA stability at postmortem 60h has decreased than at postmortem(pm) Oh. One clone of differential display(DD)-polymerase chain reaction(PCR) products more increased even at pm 60h than at pm Oh. The cDNA clones from this cloned DD-PCR product indicated 100% homology toward the cDNA clones in Gen Bank (Mus musculus LOC229510, mRNA). Conclusion: Even in postmortem time, increased gene expression could be observed.
