Summary:The article focuses on the development in the concept of fluorescence microscopy which includes stimulated emission depletion (STED) microscopy and sub diffraction resolution imaging. Sub diffraction resolution microscopy is localized by imaging with a sensitive camera that rely on photoactivatable or photoswitchable fluorophores. Engineering dark states of desired duration are allowed by controlling the photophysics.
